Summary
David Sinclair is a prominent Australian-American biologist and professor of genetics at Harvard Medical School, where he serves as the founding director of the Paul F. Glenn Laboratories for the Biological Mechanisms of Aging. He is best known for his research into the epigenetics of aging, specifically his "Information Theory of Aging," which suggests that aging is a reversible process caused by the loss of cellular identity over time. Sinclair has co-founded over 15 biotechnology companies, including Life Biosciences and Sirtris Pharmaceuticals, and authored the bestselling book "Lifespan: Why We Age – and Why We Don't Have To." His recent work involves using Yamanaka factors for epigenetic reprogramming, which led to a landmark agreement with the FDA for Life Biosciences to begin human clinical trials aimed at curing blindness by rejuvenating eye cells.

**David Andrew Sinclair** AO (born June 26, 1969) is an Australian-American biologist and academic known for his research on aging and epigenetics. Glenn Laboratories for the Biological Mechanisms of Aging at Harvard, of which Sinclair became the founding director. Sinclair's lab has continued to work on resveratrol and analogues of it as part of their research program in anti-aging. In December, 2020, Sinclair's group published that three Yamanaka transcription factors, Oct4, Sox2, and Klf4, when delivered together in a virus, could safely reverse the age of human and mouse cells, and restore the vision of old mice and mice with glaucoma. In January 2023, Sinclair's lab published research in *Cell "Cell (journal)")* purporting to support his Information Theory of Aging, the idea that mammalian aging is due to the loss of epigenetic information, and that Yamanaka factors could exert a degree of artificial control over senescence and rejuvenation in mice. Sinclair's claims of reverse aging have received criticism from other scientists. **^** "The Anti-Aging Supplements David Sinclair Takes | Skeptical Review".
- David Sinclair
He obtained his Ph.D. in Molecular Genetics at the University of New South Wales, Sydney in 1995 and worked as a postdoctoral researcher at M.I.T. with Dr. Leonard Guarente where he co discovered a cause of aging for yeast as well as the role of Sir2 in epigenetic changes driven by genome instability. The Sinclair lab was the first to identify a role for NAD+ biosynthesis in regulation of lifespan, that sirtuins activated by CR in mammals (*A Unified Theory of Caloric Restriction, 2005*), the identification of small molecules that activate SIRT1 such as resveratrol, how organisms appear to have evolved to sense plant stress and declining food supply by having plant stress metabolites such as polyphenols activate longevity defenses (*The Xenohormesis Hypothesis, 2006*), that relocalization of epigenetic factors such as SIRT1 in response to DNA breaks may be a cause of aging (*The RCM Hypothesis of Aging, 2008*), how miscommunication between the mitochondrial and nuclear genomes is a cause of age-related physiological decline (*The Mitochondrial Oasis Hypothesis, 2009*), and the idea that epigenetic changes causing cells to lose their identify are a main cause of aging and this is a reversible process (*The Information Theory of Aging, 2019*).

In this pioneering study published January 12, 2023 in Cell, Professor Sinclair and colleagues report that loss of epigenetic information causes and accelerates aging, and that these changes are reversible via epigenetic reprogramming. This finding supports Professor Sinclair's Information Theory of Aging–and the notion that aging is reversible. As detailed in this landmark study published December 2, 2020 in Nature, Professor Sinclair and colleagues have used epigenetic reprogramming to reverse vision loss in aged mice afflicted with glaucoma. In this groundbreaking book, Professor Sinclair takes the reader to the frontlines of aging research and reveals incredible breakthroughs—many from his own lab—that demonstrate how we can slow down, or even reverse, aging. ## Lifespan with Dr. David Sinclair. Essentials: The Biology of Slowing & Reversing Aging | Dr. David Sinclair Andrew Huberman 7390000 subscribers 5272 likes 186486 views 30 Oct 2025 In this Huberman Lab Essentials episode, my guest is Dr. David Sinclair, PhD, a professor of genetics at Harvard Medical School and a leading expert on the biology of aging. We discuss the cellular and molecular mechanisms of aging—and how specific behaviors, such as fasting, regular exercise and NAD⁺-boosting compounds like NMN, can activate the body's natural longevity pathways. This discussion highlights how lifestyle choices profoundly influence the aging process and may even slow or reverse key aspects of biological aging. **Who we are:** The Sinclair research group is a world leader in the understanding of why we age and how to reverse it. Our lab has trained over 100 people who will always remain part of the extended Sinclair lab family. Students and postdocs who train in our lab leave with a broad education in the latest technologies and approaches to science, along with skills in leadership and entrepreneurship, going on to successful careers in industry and academia. They are inventors on patents and some have founded successful companies out of the Sinclair lab or other labs. To date, our lab members have had a hand in the formation of over 15 companies, including Sirtris, Spotlight, Iduna, Immetas, Genocea, Life Biosciences, Jumpstart Fertility, Continuum, Arc-Bio, EdenRoc, OvaScience, Cohbar, Selphagy, Senolytic Therapeutics, and Animal Biosciences. **What we do:** Students and postdoctoral fellows in the lab can choose to learn a variety of techniques, including bioinformatics, molecular biology, epigenetic reprogramming, and animal physiology.
